Buscemi's name has Arabic origins, specifically from the word ''qalʾat ʾAbū Šāma'' meaning ''Castle of the man with the mole''. The name derives from the strategic and military position of the city, symbolized by the castle dominating the view of Anapo and the territories of the Free Consortium of Ragusa and Modica. The nickname ''of the man with the mole'' could refer to the prophet Mohammed. Over the centuries, it has been Latinized in various ways, until it arrived at its current name of Buscemi.

Buscemi is located in a central position of Sicily, 50 km west of Syracuse, 45 north of Ragusa and 70 south of Catania. The town is situated on the southern side of Mount Vignitti, which is located in the central part of the Iblei Mountains. The city is surrounded by historical sites such as ancient Akrai, Casmene and Pantalica. The nearest municipalities are Buccheri and Palazzolo Acreide, both less than ten kilometers away.

Buscemi is the thirteenth municipality of the Province of Syracuse in terms of extension of the territory with 51 km², but the inhabited center extends for just over 1 km². It is the municipality with the highest maximum altitude (987 m above sea level), the third furthest west based on the town hall, and the third with the greatest altitude range (655 m).
